HS-20093 is a novel antibody-drug conjugate (ADC) developed by Hansoh Pharmaceutical, a company located in China. It features a fully humanized anti-B7-H3 monoclonal antibody that is linked to a topoisomerase inhibitor (TOPOi) payload. This drug specifically targets B7-H3, an immunoregulatory protein that is overexpressed on both tumor and immune cells, playing a role in inhibiting T-cell activation and facilitating tumor progression. Upon administration, HS-20093 binds to B7-H3 on tumor cells, which leads to internalization and cleavage of the linker, resulting in the release of the cytotoxic payload that induces apoptosis in B7-H3-expressing tumor cells, although the precise mechanism of action remains under investigation.

Currently, HS-20093 is being assessed in Phase 2 clinical trials in China, concentrating on head and neck cancer, esophageal cancer, osteosarcoma, sarcoma, and prostate cancer.
