Acimtamig (AFM13) is a first-in-class ICEandreg; (innate cell engager) molecule developed by Affimed, designed to target CD30 on cancer cells and CD16A on innate immune cells. This innovative therapy aims to leverage the body’s immune system to effectively target and kill cancer cells in CD30-positive lymphomas. Acimtamig represents a novel approach to immunotherapy, harnessing the power of innate immune cells, specifically natural killer (NK) cells, to induce a potent anti-tumor response. It has shown significant promise as a monotherapy in clinical trials, demonstrating meaningful antitumor activity in patients with CD30-positive lymphomas.

One of the most striking results of acimtamig has been its combination with anti-PD-1 antibodies in Hodgkin lymphoma. In clinical studies, the combination therapy led to an overall response rate (ORR) of 88% at the highest treatment dose, with a complete response (CR) rate of 46%, as assessed independently. These outcomes were superior to the results observed with anti-PD-1 therapy alone, highlighting acimtamig's potential to enhance the efficacy of existing treatments and offer a more effective option for patients with Hodgkin lymphoma.
Further studies have demonstrated even more compelling results when acimtamig was combined with fresh, cord blood-derived NK cells. In a Phase 1/2a study involving heavily pretreated patients with Hodgkin lymphoma and Non-Hodgkin lymphoma, acimtamig produced unprecedented outcomes. In a cohort of 32 Hodgkin lymphoma patients treated with the recommended Phase 2 dose, the ORR was 97%, and the CR rate was 78%. These patients also experienced a median event-free survival (EFS) of 9.8 months, with 84% alive at 12 months. The median duration of response (DoR) was 8.8 months, demonstrating the lasting benefits of this combination. Moreover, the treatment showed a favorable safety and tolerability profile.
Affimed is now expanding on these promising findings in its Phase 2 study, LuminICE-203, which explores the combination of acimtamig with alloNKandreg;, an off-the-shelf, allogeneic NK cell product from Artiva Biotherapeutics. This study focuses on patients with relapsed/refractory classical Hodgkin lymphoma (r/r cHL) and includes an exploratory arm for relapsed/refractory peripheral T-cell lymphoma (r/r PTCL). With these ongoing studies, acimtamig has the potential to become a transformative therapy for patients with CD30-positive lymphomas.
